چکیده

The importance of the renin-angiotensin system in the development and maintenance of one-kidney renal hypertension in the dog was assessed by chronically inhibiting angiotensin by continuous infusion of [Sar'-,Ala]angiotensin II or a converting enzyme inhibitor (SQ 20881). Angiotensin blockade was begun 1-2 days before renal artery constriction and continued for 6-7 days throughout the high rerun phase of hypertension. Chronic hypertension was produced despite continuous angiotensin inhibition with [Sar',Ala]angiotensin II or SQ 20881. Mean arterial pressure rose to hypertensive levels during the |Sar,Ala"]angiotensin II infusion, but the increase occurred more slowly than when the renin-angiotensin system was intact. Although chronic infusion of the angiotensin II analogue itself produced no measurable vasopressor agonism, the analogue appeared to be mildy agonistic for the kidney and adrenal cortex. Chronic SQ 20881 infusion consistently lowered mean arterial pressure by about 10 mm Hg in unilaterally nephrectomized normotensive dogs and potentiated the vasodepressor action of bradykinin. Renal artery constriction during the SQ 20881 infusion for 6 days produced a gradual increase in mean arterial pressure (18 mm Hg above the control level); discontinuation of SQ 20881 was followed by a further elevation in arterial pressure. The present observations demonstrate that chronic hypertension developed and was sustained in spite of minimal or no activation of the renin-angiotensin system during the acute high renin phase of one-kidney hypertension.

Attenuation of nephrotoxic acute renal failure in the dog with angiotensin-converting enzyme inhibitor (SQ-20,881).

Angiotensin-converting enzyme inhibitor was used in dogs with uranyl nitrate-induced acute renal failure to evaluate (1) a possible protective effect of angiotensin blockade and (2)the role of angiotensin II in the generation of renal failure in this model. Angiotensin-converting enzyme inhibitor treatment attenuated the fall in glomerular filtration rate and renal blood flow during the first 6...

بررسی پاسخدهی عروق مزانتر به آنژیوتانسین I و II در خلال ایجاد پرفشاری خون دو کلیه ای گلدبلات و اثر کاپتوپریل بر آن

Essential hypertension is one of the risk factors of cardiovascular diseases. Hypertension etiology is not completely known, it seems that rennin-Angiotensin system has an important role in its etiology, Thus better recognition of this system and its activity changes or vascular reaction changes to different parts of this system during progressive hypertension can be more effective in better re...

Crocin prevents acute angiotensin II-induced hypertension in anesthetized rats

Objective: Angiotensin II (Ang II), the main product of renin-angiotensin system (RAS) has a well-known role in cardiovascular regulation. Over-production of Ang II is one of the important underlying mechanisms of hypertension. In this study, the effect of crocin on cardiovascular responses in rats with acute hypertension induced by Ang II was evaluated. Materials and methods: Rats were divided...

Intrarenal renin-angiotensin-sodium interdependent mechanism controlling postclamp renal artery pressure and renin release in the conscious dog with chronic one-kidney Goldblatt hypertension.

We studied postclamp renal artery pressure and renin release in eight conscious dogs with one-kidney Goldblatt hypertension. On normal sodium intake, intrarenal blockade of angiotensin II with Sar1-Ala8-angiotension II (P-113, saralasin acetate) markedly decreased postclamp renal artery pressure and increased renin release during the first 5 days after renal artery constriction. We found that 1...

Blockade of Central Angiotensin II AT1 Receptor Protects the Brain from Ischemia/Reperfusion Injury in Normotensive Rats

Background: Stroke is the third leading cause of invalidism and death in industrialized countries. There are conflicting reports about the effects of Angiotensin II on ischemia-reperfusion brain injuries and most data have come from chronic hypertensive rats. In this study, hypotensive and non-hypotensive doses of candesartan were used to investigate the effects of angiotensin II AT1 receptor b...
